First your console should have CFW installed to play PS2 ISO.

After you have copied the PS2 ISO to /dev_hdd0/PS2ISO, you need to mount the ISO using a backup manager like webMAN MOD, IRISMAN, multiMAN or ManaGunZ.

If you encrypt the ISO, you can create a PKG and it will launch directly when you start the game from XMB.

Yeah with a super slim you need to encrypt your PS2 dump in order for it to work in .BIN.ENC format but at least Apollo save tool helps with that. While on my non backwards capable FAT PS3 I am able to run unencrypted ISO. Glad I swapped HDDs with my super slim and immediately dumped the root key of that CFW compatible console.
